At the moment several manufacturers offer 320GB. These are standard size (3/8" / 9.5mm thick) and are easily available. A 500GB 2.5" HDD is due from Hitachi in the near future but it remains to be seen how soon it reaches the retail chain. However, it is 1/2" / 12.7mm thick so it will not fit many notebooks. Samsung have promised a 500GB 3/8" thick HDD for mid year, but there's no additional info.

Right now the highest HD capacities are the 320GB 5400RPMs, but that will change in the near future.

All you have to do is dismantle the case, which many have easily done.

I just use 2 x 200GB 7200rpm HDD's in RAID 0.
I believe 200GB is the biggest you can get with a spindle speed of 7200rpm. Any higher and the spindle speed will drop to 5400rpm.
